Best: The running joke about old-timey photography
While I wish more of the film had played off of the authentically weird ways people did things in the Old West, the running joke about how no one smiles for photographs paid off for me every time it came back up. There's something genuinely ridiculous about trying to imagine anyone holding a smile for ten solid minutes, and I love the final gag. To me, this points at what could have been the movie's biggest strength if they'd done more of it, taking a clear-eyed look at the weirdest details of a time and place.
